Eli Lilly Study of the LY3954068 (Anti-Tau siRNA) Drug

The main purpose of this study is to evaluate the safety of LY3954068 in participants with early symptomatic Alzheimer’s Disease (AD). The study will also investigate how much LY3954068 gets into the bloodstream and will test the effects of LY3954068 on markers of AD.

Study Timeline

• Single intrathecal injection
• 5 Lumbar Punctures
• Minimum of 2-night stay in the Duke Early Phase Research Unit

Inclusion Criteria

• Age 50 – 85
• Mini-mental state examination of 18 – 30
• BMI of 18 – 40
• Medical co-morbidities must be stable with no medication changes within the past 3 months.
• Amyloid pathology is not required.

Exclusion Criteria

• Anti-tuberculosis therapy within the past 12-months
• Significant lumbar pathology that would interfere with lumbar punctures
• More than 2 areas of superficial siderosis
• More than 5 micro hemorrhages, or a prior history of macro hemorrhages
• Systemic anticoagulants or antithrombotic (low dose acetylsalicylic acid (ASA) is allowed)
• Significant renal impairment (estimated glomerular filtration rate of less than 45)
• Platelet count of less than 100,000
